Hyaluronoglucosaminidase (hyaluronidases, EC 3.2.1.35) catalyzes the hydrolysis of hyaluronic acid (HA), a high molecular weight non-sulfated linear glycosaminoglycan, which is composed of repeating disaccharide units, D-glucuronic acid (GlcA) and N-acetyl glucosamine (GlcNAc). The enzyme cleaves the beta-1,4-glycosidic linkage between N-acetyl-glucosamine and glucuronate within the HA chain, producing non-reactive oligomeric end products. They also hydrolyses 1,4-beta-D-glycosidic linkages between N-acetyl-galactosamine or N-acetylgalactosamine sulfate and glucuronic acid in chondroitin, chondroitin 4- and 6-sulfates. To a small extent, they hydrolyze dermatan sulfate. Hyaluronidases are wide spread in nature, being found in mammals, insects, leeches, and bacteria. The diversity allows them to have different catalytic mechanisms, substrate specificities, affinities to inhibitors, and notably, different pH optima.

Hyaluronoglucosaminidases have different roles in different species. In venoms of various organisms, such as those of bees, wasps, spiders, scorpions, fish, snakes and lizards, hyaluronidases enhance the noxious action of substances by helping to spread constituents of the venom cocktail at the injection site. In humans, increased levels of hyaluronidase are found in some cancers, such as colorectal, bladder, prostate, and breast cancer. Therefore, the enzyme is used as a marker for the diagnosis of growing tumors. The enzyme is being heavily studied for its versatile functions.
