Enzyme Activity Measurement of Non-Reducing End Alpha-L-Arabinofuranosidase

Non-reducing end alpha-L-arabinofuranosidase (Abf, EC 3.2.1.55) are exo-acting enzymes that hydrolyze alpha-(1→2), alpha-(1→3) and alpha-(1→5) glycoside bonds and release arabinofuranosyl moieties from a wide variety of arabinose-containing substrates, including arabinan, arabinogalactans, heteroxylans, and synthetic compounds such as p-nitrophenyl-alpha-L-arabinofuranoside (pNP-α-L-Araf). According to the glycoside-hydrolase classification system, Abfs are present in six families (GH-3, 43, 51, 54, 62 and 93). GH-51 and GH-54 are the two major families, containing almost exclusively Abfs.

Alpha-L-arabinofuranosidases attract an increasing attention due to the biotechnological applications in ethanol production for biofuels and in the food and feed industries. Abfs catalyze the enzymatic hydrolysis of heteroxylans, the most common hemicelluloses and complex polysaccharides, which are composed of the beta-D-xylose backbone with possible substituents of various neutral and acidic sugars as well as acetyl groups and phenolic acids. Hemicelluloses are key structural components of plant cell walls and are regarded as the second most abundant source of renewable carbon after cellulose. However, the complicated structure limits the efficient utilization of hemicelluloses. The ability of Abfs in hydrolyzing hemicelluloses makes them widely used in the degradation of biomass such as lignocellulose, hemicellulose, and pectin. In agriculture, Abfs are used to improve the digestibility of animal feed. In addition, Abfs can be used in the analysis, paper production, and synthesis of chemicals. With the increasing studies on a variety of applications of Abfs, seeking high catalytic activity is the essential topic.
