Alpha-amylases (1,4-R-glucan 4-glucanohydrolase, EC 3.2.1.1) are ubiquitous enzymes which catalyze the breakdown of amylose and amylopectin through the hydrolysis of internal alpha-1,4-glycosidic linkages with net retention of anomeric configuration, simultaneously with the production of glucose and maltose. These enzymes act on starch, glycogen and related polysaccharides and oligosaccharides in a random manner. It is the major form of amylase in human and mammals whereas in fungi this enzyme is extracellularly secreted.
Alpha-amylases have been exploited for several applications including the food industry, drug development, and analysis. In the food industry, alpha-amylase is an important industrial enzyme in brewing and alcohol production. In addition, it is utilized in high-fructose syrup production. In clinical studies, the inhibition of alpha-amylase can significantly reduce the post-prandial increase of blood glucose and therefore can be an important strategy in the management of blood glucose level in type 2 diabetic and borderline patients. In the analysis, the alpha-amylase from A. amylolyticus is shown to be used in a sensitive method to detect very low concentration of heavy metals in drinking water. The activity determination methods of alpha-amylase differ in these applications. The commonly used methods are starch iodine test and the Phadebas amylase test, which is the qualitative and quantitative test employed in many industries.
