Phosphogluconate Dehydrogenase (NADP+-dependent, decarboxylating) is the second dehydrogenase of the pentose phosphate pathway (PPP). The pathway mainly contains two parts: an oxidative branch and a non-oxidative, rearrangement branch. The pathway is present in most species and most tissues. The main function of PPP is to produce NADPH and pentose for biosynthetic reactions. NADPH is essential in protecting the organism against oxidative stress and is required for a variety of reductive biosynthetic reactions, particularly lipid production. 6PGDH plays a crucial role in this pathway because it is the very enzyme that produces NADPH along with the production of ribose 5-phosphate, which is the source of nucleotide and nucleic acid synthesis. Due to the important functions it plays in organisms, this enzyme may pave the path to the treatment of some diseases. For example, it has been posited as a potential drug target for African sleeping sickness (trypanosomiasis).
