Enzyme Activity Measurement of Isocitrate Dehydrogenase (NADP+) Using Spectrophotometric Assays

Isocitrate dehydrogenase (ICDH) is an enzyme in the tricarboxylic acid (TCA) cycle that catalyzes the dehydrogenation and the subsequent decarboxylation of isocitrate to α-ketoglutarate and CO2. EC 1.1.1.42 is a NADP+-linked enzyme that is found in both mitochondria and cytoplasm. The enzyme from some species can also use NAD+ but much more slowly. Isocitrate dehydrogenase (NADP+) requires Mn2+ or Mg2+ for activity. Unlike EC 1.1.1.41 (isocitrate dehydrogenase (NAD+)), oxalosuccinate can be used as a substrate of this enzyme. Isocitrate dehydrogenase (NADP+) catalyzes the important step in the tricarboxylic acid (TCA) cycle. IDH1 and IDH2 are two isoforms of isocitrate dehydrogenase (IDH) in human using NADP+ as the cofactor. These two enzymes are of great clinical significance. Specific mutations in IDH1 have been found in several brain tumors including astrocytoma, oligodendroglioma, and glioblastoma multiforme. Due to the important role in metabolism and the pivotal value in medical research, more and more studies have started on this enzyme.
