Testosterone 17-beta-dehydrogenase (NADP+) (EC1.1.1.64) is an enzyme that catalyzes the oxidation of testosterone, using NADP+ as the redox cofactor, to produce androst-4-ene-3,17-dione. This enzyme exists in many mammals, such as Oryctolagus cuniculus, Homo sapiens, Sus scrofa, and Rattus norvegicus. The enzyme is a member of the oxidoreductases family, more specifically, the oxidoreductases that catalyze the CH-OH group of the substrate and use NAD+ or NADP+ as the cofactor. The systematic name of this enzyme class is 17beta-hydroxysteroid: NAD+ 17-oxidoreductase, which is also called 17-ketoreductase, NADP-dependent testosterone-17beta-oxidoreductase, or testosterone 17beta-dehydrogenase (NADP). Note that testosterone 17-beta-dehydrogenase (NADP+) is involved in the steroid hormone biosynthesis pathway and some other metabolic pathways.

Testosterone 17-beta-dehydrogenase (NADP+) plays an important role in the pathway of androgen biosynthesis, which is part of the super pathway of steroid hormone biosynthesis. This pathway constitutes the main reactions of the conversion of the inactive C21 steroid hormone precursor pregnenolone to the androgens: androstenedione, testosterone and 17β-hydroxy-5α-androstan-3-one. Mutations in the enzymes in this pathway result in varying degrees of sexual and genital malformation. Testosterone is the input to estradiol biosynthesis I (via estrone), and it is also the output of the reaction which catalyzed by testosterone 17-beta-dehydrogenase (NADP+) to convert androst-4-ene-3,17-dione. Testosterone is the key male sex hormone, and it regulates a number of processes in the male body. It was proved by many studies that testosterone can be correlated with obesity and heart diseases. Therefore, testosterone 17beta-dehydrogenase(NADP+) would be a promising target of potential disease therapies.
