Glucan 1,4-alpha-maltohydrolase (maltogenic amylase, EC 3.2.1.133) is a potential catalyst for malto-oligosaccharide production. It is an amylolytic enzyme from glycosyl hydrolase family 13 (GH13). The enzyme catalyzes the hydrolysis of 1,4-alpha-D-glucosidic linkages in polysaccharides so as to remove successive alpha-maltose residues from the non-reducing ends of the chains. In addition, maltogenic amylase also acts on starch and related oligosaccharides.

Oligosaccharides have attracted much attention due to their numerous health benefits and the improvement of the physicochemical properties of foods. Therefore, a progressive increase in the demand for oligosaccharides has promoted the need for efficient catalysis in production. Oligosaccharides can be synthesized chemically, but enzyme catalyzed hydrolysis is essential to many important industrial reactions, with enzymatic hydrolysis being the basis of both the liquefaction and saccharification progresses during the production of high-glucose syrups from corn starch. Hence, alpha-amylases family members are preferred in the industries such as textile and detergent field, including maltogenic amylase. Besides, this enzyme has applications in medicine modifications. For example, maltogenic amylase is utilized to increase the solubility of puerarin by transglycosylating, thus, to become a therapeutic agent of coronary heart disease, cardiac infarction, and alcoholism. The enzyme activity can be measured by the dinitrosalicylic acid (DNS) method, which relies on precise spectrophotometers.
