Sphingomyelin phosphodiesterase (SMase, EC 3.1.4.12) has been found in several microorganisms and mammals. It catalyzes the breakage of phosphodiester bond of sphingomyelin, yielding the ceramide and the water-soluble phosphate ester. However, SMase does not cleave the phosphodiester bond of glycerophospholipids and has very little activity on phosphatidylcholine. SMase requires Mg2+ for sphingomyelin hydrolytic activity.
Eukaryotic SMases have been classified according to their pH optima and are known as acid SMase, alkaline SMase, and neutral SMase. Among these types, neutral sphingomyelinase is most studied. In mammalians, neutral SMase is thought to play a key role in sphingolipid metabolism and there is increasing evidence implicating sphingomyelin metabolites in cell signaling, cell proliferation, and apoptosis. Thus, sMase has vital research significances in pathology, diagnostic and drug development. In diagnostic, this enzyme becomes a potential biomarker for many diseases, for example, the sphingomyelinase activity in fecal reflects the human intestinal mucosa enzyme level and may indicate colorectal adenocarcinoma. In drug development, inhibiting or stopping neutral SMase activity is an important strategy to prevent neuron death from ischemia. Besides, this enzyme is a potential target in the treatment of allergic reactions. Although the crystal structure of human sphingomyelin phosphodiesterase has not been solved yet, more and more studies are underway because of its huge potentials in clinical treatment.
