Enzyme Activity Measurement for Deoxyribonuclease I

Deoxyribonuclease I (DNase I, EC 3.1.21.1) is a glycoprotein. It acts on double-stranded DNA and catalyzes the endonucleolytic cleavage to 5'-phosphodinucleotide and 5'-phosphooligonucleotide end-products. DNase I is a divalent-cation dependent enzyme which exhibits optimal activity in the presence of Ca2+ and Mg2+ or Mn2+. The enzyme has been used as a biochemical probe in a number of experiments and is now an established tool in molecular biology.

In addition to the applications in biological research, DNase I also has important clinical significances. It has been suggested to be one of the deoxyribonucleases responsible for DNA fragmentation during apoptosis. DNase I is correlated with several diseases. For example, abnormal activity of DNase I is found in patients with type 2 diabetes as well as the increasing expression of DNase I in the pancreas of diabetic patients. Other diseases include systemic lupus erythematosus, chronic pancreatitis, stomach cancer, and glomerulonephritis. Therefore, DNase I is regarded as a diagnostic marker for the early detection of diseases. Simple detection methods and accurate results are indispensable for activity measurement. The assay of DNase needs some precautions, such as the choice of buffer, the utilization of proteases, the concentration of SDS and guanidine hydrochloride, and the freezing-thawing process.
