Carboxylesterases (EC 3.1.1.1; carboxylic-ester hydrolase) are metabolic enzymes that are found across all kingdoms. Most of these enzymes are part of the α/β hydrolase fold superfamily of proteins. These enzymes catalyze the hydrolysis of carboxylic esters into their corresponding alcohols and carboxylic acids. Members of the α/β hydrolase fold superfamily share a conserved core structure. Carboxylesterases contain a catalytic triad consisting of a serine, an acidic amino acid, and a histidine. Mutation of any one of these residues results in a non-functional enzyme. The catalytic triad forms a charge relay system to create a nucleophilic serine that can attack the ester substrate. Carboxylesterases have broad substrate specificity. Several highly homologous carboxylesterase isozymes that exhibit overlapping substrate specificities have been identified in rodents. Importantly, carboxylesterases from microsomes also catalyze the reactions that are naturally catalyzed by many other hydrolases.
