The aldo-keto reductases make up a superfamily of NAD(P)-dependent oxidoreductases with broad physiological roles. These enzymes show substantial diversity in terms of substrate preferences; each member of this superfamily catalyzes the reduction of a range of aliphatic and aromatic aldehydes and ketones, including sugars, steroids, and xenobiotics. Succinic semialdehyde reductase belongs to aldo-keto reductase family 7 (AKR7) and catalyzes the NADP+-dependent oxidation reaction of 4-hydroxybutanoate to produce succinate semialdehyde. In human, this enzyme is encoded by AKR7A2. Up to now, some other members of the AKR7 family have also been cloned, including human AKR7A3, rat AKR7A4, and mouse AKR7A5. These enzymes have broad substrate specificity and may have NADPH-dependent aldehyde reductase activity towards 2-carboxybenzaldehyde, 2-nitrobenzaldehyde and pyridine-2-aldehyde, as indicated by in vitro tests. There is mounting evidence showing that human AKR7A2 can function as a succinic semialdehyde (SSA) reductase in brain and may play a role in the synthesis of γ-hydroxybutyrate (GHB). So far, only succinic semialdehyde reductase in mouse has been well studied. This enzyme has a typical AKR7 family local structure, common to all the members of the aldo-keto reductase superfamily. Due to the specific catalytic chemistry and their promiscuity of the substrate, these enzymes have huge potentials in food processing, fine chemicals, and pharmaceutical industries. Therefore, more and more researchers have turned their research topics to these enzymes in order to fill the blanks related to its characteristics.
