Mevaldate reductase was first found related to mevalonate biosynthesis in rat liver. The high activity of this enzyme helped early workers in the field to obtain a high incorporation rate of labelled mevaldic acid into non-saponifiable lipids in rat liver homogenates, which led to the theory that free mevaldate is an intermediate in sterol biosynthesis. Studies on this enzyme shows it is inhibited by barbiturates. Sodium aminobarbitone and thiobarbituric acid are two main inhibitors of this enzyme. The properties of this enzyme is similar to those of other animal tissue aldehyde reductases (EC 1.1.1.2), and it is suggested that mevaldate reductase be reclassified as one of this group. To assay the activity of this enzyme, the best and efficient way is to employ the spectrophotometric assay method. According to the basic principle of spectrophotometric absorption, measurement of mevaldate reductase activity can be carried out by detecting the consumption of NADP+ or the generation of NADPH.
