Linoleate 13S-lipoxygenase (EC 1.13.11.12, formerly EC 1.13.1.13 and EC 1.99.2.1.) belongs to the lipoxygenases family which act on polyunsaturated fatty acids in lipids. These enzymes are responsible for a wide range of essential functions in eukaryotes, while the biological roles of prokaryotes lipoxygenases remain unclear. In mammals, lipoxygenases produce leukotrienes and lipoxins, which constitute an important family of signaling molecules that affect a number of biological processes, such as asthma, atherosclerosis, and cancer. Linoleate 13S-lipoxygenase specifically oxidizes linoleate and alpha-linolenate, the two most common polyunsaturated fatty acids in plants, by inserting molecular oxygen at the C-13 position with (S)-configuration. The conserved structures contain non-heme iron, although some fungal enzymes contain manganese instead of iron.
This enzyme plays an important role in plants for the participation of biosynthesis pathways, such as divinyl ether biosynthesis II and jasmonic acid biosynthesis. The former has been reported related to the defense machinery in some plants. The latter is involved in the regulation of abiotic or biotic stresses responses during the plant growth and development. Besides, it also responses to wounding of plants, systemic acquired resistance, and pest control. Meanwhile, linoleate 13S-lipoxygenase takes part in alpha-linolenic acid metabolism and linoleic acid metabolism, which produces precursors for several important compounds. Although the crystal structure of this enzyme from human sources has not been identified, linoleate 13S-lipoxygenase was unambiguously shown to have a protumorigenic role in prostate cancer. Due to its key position in organism growth and metabolism, and its significances in disease treatment, an explosion of studies on this enzyme can be foreseen in the near future.
