D-lactate dehydrogenases and L-lactate dehydrogenases (D-LDH and L-LDH) reduce pyruvate to D- and L-lactate, respectively, with oxidization of NADH into NAD+, and act as the last step of the glycolytic pathway under anaerobic conditions. D-lactate dehydrogenase is also known as:
D-LDHs belongs to a large D-2-hydroxyacid dehydrogenase (D-HydDH) superfamily, which comprises various types of D-isomer dehydrogenases, such as the D-glycerate, D-3-phosphoglycerate, D-2-hydroxyglutarate and D-2-hydroxyisocaproate dehydrogenases. Although these enzymes bear highly divergent primary sequences, their tertiary structures are quite similar to one another. The Lactobacillus D-LDHs consistently have a dimeric structure comprising identical subunits, and exhibit hyperbolic shaped saturation curves for substrate pyruvate. And in E. coli cells, D-LDH gives a homotetrameric structure. Bacterial allosteric D-LDHs are commonly activated by FBP (fructose 1,6-bisphosphate) and the inhibition occurres when the oxmate presents in the solvent. D-lactate dehydrogenases activity was determined by measuring the variation of the absorbance at 340 nm derived from NADH.
