UDP-glucose-hexose-1-phosphate uridylyltransferase (EC 2.7.7.12) is an enzyme that catalyzes the reversible transfer of the uridine 5’-phosphoryl moiety from uridine 5’-diphosphate glucose (UDP-Glc) to galactose-1-phosphate (Gal-1-P) to produce uridine 5’-diphosphate galactose (UDP-Gal) and glucose-1-phosphate (Glc-1-P). The reaction is part of the Leloir pathway of galactose metabolism required for the normal equilibration of UDP-hexoses among most organisms. In this pathway, galactose is first phosphorylated to Gal-1-P by galactokinase. UDP-glucose-hexose-1-phosphate uridylyltransferase then catalyzes the transfer of the uridylyl group from UDP-Glu to Gal-1-P to produce UDP-Gal and Glc-1-P, which then enters the glycolytic pathway. UDP-glucose-hexose-1-phosphate uridylyltransferase is apparently ubiquitous, and it is a member of the histidine triad (HIT) superfamily, which functions as either nucleotide hydrolase or transferase that reacts with the α-phosphorus of nucleotide.
It is noteworthy that galactosemia, the human autosomal recessive genetic disease, results from the impairment of UDP-glucose-hexose-1-phosphate uridylyltransferase activity, leading to toxic levels of galactose and Gal-1-P. Therefore, UDP-glucose-hexose-1-phosphate uridylyltransferase may play a pivotal role as a target for the inhibitor design in galactosemia treatment.
