Sugar-phosphatase (EC 3.1.3.23; sugar-phosphate phosphohydrolase) is an enzyme that catalyzes the hydrolysis of the phosphate esters of sugars. Sugar-phosphatase is located exclusively in the periplasmic space membrane of bacteria. This enzyme can be purified from various bacteria, such as Escherichia coli, Neisseria meningitidis, and Lactococcus lactis. The sugar-phosphatases from E. coli and N. meningitidis are very similar to each other in terms of substrate specificity, intracellular localization, effects of glucose on enzyme biosynthesis, and physicochemical and catalytic properties. Sugar-phosphatase belongs to the family of hydrolases, specifically those acting on phosphoric monoester bonds. Note that this enzyme has a wide substrate specificity, which can act on aldohexose 1-phosphates, ketohexose 1-phosphates, aldohexose 6-phosphates, ketohexose 6-phosphates, both phosphate ester bonds of fructose 1,6-bisphosphate, phosphoric esters of disaccharides, and pentose and triose phosphates.
Sugar-phosphatase may be a unique homologous enzyme present in certain microorganisms which share a common system of transport and metabolism of sugars and phosphates. Besides, sugar-phosphatase may be the enzyme exploited for the cleavage of phosphorylated sugars transported via either the phosphoenolpyruvate-hexose phosphotransferase or other systems. Thus, to understand the physiologic role of this enzyme and its relationship with the phosphoenolpyruvate-hexose phosphotsansferase system, it is indispensable to be able to detect and monitor the activity of sugar-phosphatase.
