Phosphodiesterase I (5'-NPDase, EC 3.1.4.1) acts on both ribonucleotides and deoxyribonucleotides. It hydrolytically removes 5'-nucleotides successively from the 3'-hydroxyl termini of 3'-hydroxy-terminated oligonucleotides. However, 5'-NPDase has low activity towards polynucleotides and the hydrolysis will be inhibited if there is a 3'-phosphate terminus on the substrate. Besides, 5'-NPDase is inhibited by reducing agents such as glutathione, cysteine, and ascorbic acids. The enzyme requires Mg2+ for its maximum activity.

5'-NPDase is a glycoprotein that forms disulfide-bonded homodimers in the plasma membrane and mineral-depositing matrix vesicles of osteoblasts and chondrocytes. This enzyme has extensive applications. In molecular biology, the enzyme has been widely utilized as a tool for structural and sequence studies of nucleic acids. It is also a useful enzyme for direct detection of certain tandem DNA damage. In addition, this enzyme has important positions in medicine and drug development. 5'-NPDase is a potential target for drug design in the treatment of calcium pyrophosphate dehydrate deposition disease as well as cancer. It contributes to the pharmacology research because 5'-NPDase plays an important role in regulating cAMP and cGMP, and thus becomes an important site for the pharmacological intervention, such as the inhibition studies for Parkinson's disease. Besides, this enzyme has huge potentials in the biosynthesis of 5’-mononucleotide.
