UTP-monosaccharide-1-phosphate uridylyltransferase (PsUSP, EC 2.7.7.64) is a unique nucleotide-sugar pyrophosphorylase that can utilize diverse sugar 1-phosphates to form their corresponding UDP-sugars. The available substrates include glucose 1-phosphate, galactose 1-phosphate, glucuronic acid 1-phosphate, and xylose 1-phosphate, whereas mannose 1-phosphate, L-fucose 1-phosphate, and glucose 6-phosphate do not serve as substrates. The enzymatic reaction is limited to using UTP rather than other nucleotide triphosphates. PsUSP requires Mg2+ or Mn2+ for maximal activity.
PsUSP is important for the biosynthesis of activated carbohydrates which play pivotal roles in a vast amount of cellular processes. Activated carbohydrates participate in glycoprotein biosynthesis, cellular responses, signal transductions, and sucrose and polysaccharide metabolisms in plants. Moreover, they are related to the surface modification of cell walls or even the complete disguise of organisms by forming a complex protective glycocalyx coat. Such camouflage strategy contributes to escaping the recognition by an infected host. Every single year, approximately 12 million people are infected by protozoan parasites and 60,000 people died of it. Therefore, PsUSP has become an attractive target for the development of anti-parasite drugs. The published crystal structure of PsUSP from Leishmania major provides an insight into the catalytic mechanism. Based on the continuous studies on this enzyme, the efficient medicines are hopefully to be developed.
