Sterol esterase (EC 3.1.1.13) has broad substrate specificity. They act on esters of sterols and long-chain fatty acids, and may be also involved in the esterification of sterols. This enzyme is activated by bile salts, therefore, it is also called bile salt activated lipase.
Sterol esterase plays important roles in the dietary uptake. This enzyme is secreted from the pancreas into the intestinal tract, where its major physiological function presents lipolytic activity. In addition, it is the only known intestinal lipase that hydrolyzes cholesteryl ester, fat-soluble vitamins, and other fatty acid esters. The deficiency of sterol esterase activity may arouse a reduced uptake of cholesteryl ester, indicating the enzyme is responsible for mediating intestinal absorption of cholesteryl esters. The lack of sterol esterase activity also causes incomplete digestion of milk fat and lipid accumulation in the ileum. Besides its roles in fat digestion, this enzyme is also implicated in the cycle of cellular homeostasis of cholesterol, which is particularly affected in tumor cells. Noticing the potentials in clinical treatment, many studies are carried out focusing on the medicine development. The screening of effective enzyme inhibitors is the vital part of the development of potential therapeutics. More specifically, theses inhibitors may be used to limit cholesterol absorption and hypercholesterolemia. In order to better characterize the effects of the inhibitors, proper activity assay methods must be developed first.
