Phosphogluconate 2-dehydrogenase, the third enzyme of the pentose phosphate pathway, catalyzes the oxidative decarboxylation of 6-phosphogluconate (6PG), giving ribulose 5-phosphate as the product, along with the reduction of NADP+ to NADPH and the release of CO2. The enzyme catalyzes the reaction via three steps: (1) oxidation of the C3-hydroxyl group of 6PG to a C3-keto intermediate, (2) decarboxylation to give 1,2-enediol and (3) simultaneous tautomerization to Ru5P. This enzyme participates in the pentose phosphate pathway. This pathway is a metabolic pathway parallel to glycolysis. Phosphogluconate 2-dehydrogenase catalyzed reaction generates reducing equivalents, in the form of NADPH through oxidation, which then provides the reducing power for the synthesis reaction in cells, such as the synthesis of fatty acids. This pathway also produces ribose 5-phosphate (R5P) and erythrose 4-phosphate (E4P), which provide materials for the synthesis of many biological building blocks, such as nucleotides. This enzyme is also involved in glutathione metabolism. Glutathione is an important antioxidant, which is capable of preventing damages to important cellular components from reactive oxygen species, such as free radicals, peroxides, lipid peroxides, and heavy metals.
