Enzyme Activity Measurement of Oligosaccharide Reducing-End Xylanase

Glycoside hydrolases (GHs) are classified into more than a hundred families based on amino-acid sequence identity. Oligosaccharide reducing-end xylanase belongs to the GH8 family owing to its similarity to GH8 endo-xylanases. Oligosaccharide reducing-end xylanase (Rex, EC 3.2.1.156) is a unique exo-oligoxylanase that hydrolyzes xylo-oligosaccharide at their reducing ends to release xylose and leaving the new reducing end in the alpha configuration. It is specific for the beta anomers of xylooligosaccharides whose degree of polymerization is equal to or greater than 3. Up till now, Rex is the only complete exo-hydrolytic enzyme that releases monosaccharide from the reducing end.

Rex is originally isolated from the bacterium Bacillus and only the crystal structure from Bacillus halodurans has been identified. In this organism, the enzyme is considered to play a key role in the intracellular xylan metabolism by cleaving xylooligosaccharides. Because Rex is an inverting enzyme that produces the alpha-anomer at the reducing end, spontaneous mutarotation of the substrate is required for subsequent processing in vitro. It remains unclear whether there are any factors supporting the mutarotation of xylooligosaccharides in vivo or if there are any biological reasons for such an inefficient processing mechanism. Therefore, the studies are focused on the basic functional research. To this end, Creative Enzymes support with the excellent activity assay service.
