Enzyme Activity Measurement of N-Acylmannosamine 1-Dehydrogenase Using Spectrophotometric Assays

N-acylmannosamine 1-dehydrogenase belongs to the SDR (short-chain dehydrogenase/reductase) superfamily, and it catalyzes a rare NAD+-dependent oxidation reaction of ManNAc (N-acetyl-Dmannosamine) and turns it into N-acetylmannosamino-lactone, which spontaneously hydrolyzes into N-acetylmannosaminic acid. This enzyme hasn’t been found in other organisms than Flavobacterium sp, and it is so far the only enzyme that catalyzes the reaction. The enzyme is also called by other terms:

  • N-acylmannosamine dehydrogenase;
  • N-acetyl-D-mannosamine dehydrogenase;
  • N-acyl-D-mannosamine dehydrogenase

N-acylmannosamine 1-dehydrogenase is involved in sialic acid catabolism in bacteria. The most common form of sialic acid is Neu5Ac (N-acetylneuraminic acid), which is considered as a biomarker for some tumoral and inflammatory states, rendering its quantification in serum important. In microgranisms, Neu5Ac splits to pyruvate and ManNAc. Usually, biological processes converse this intermediate into fructose 1,6-bisphosphate through a series of enzymes catalyze. However, there is a unique pathway exists in a soil bacterium Flavobacterium sp. 141-8, which is correlated to its ability to grow on ManNAc as the sole carbon source. The unique enzyme is N-acyl-D-mannosamine dehydrogenase, which has not been found in any other microorganism. Little studies on this unique pathway has been done, and the consecutive reactions following N-acetylmannosaminic acid still remain unknown. Thus, scientists are keen to understand this enzyme and pathway. This enzyme is currently sold commercially as a part of the sialic acid assay kit for clinical testing. While many other potential applications awaits until the enzyme is better studied and characterized.
