Malate dehydrogenase (decarboxylating) activity was first isolated from pigeon liver and has since been found in most living organisms, from bacteria to humans. Malate dehydrogenase (decarboxylating) (EC 1.1.1.39) catalyzes the reversible oxidative decarboxylation of L-malate to pyruvate and CO2 with the concomitant reduction of the cofactor NAD+ or NADP+. The reaction generally proceeds in two steps – dehydrogenation (oxidation) of malate to produce the oxaloacetate intermediate, and decarboxylation to produce pyruvate – but, interestingly, the enzyme does not decarboxylate oxaloacetate that is added exogenously.
