Glyoxylate reductase (EC 1.1.1.26) was first isolated from spinach leaves. The alternative names of this enzyme are as follows:

Glyoxylate reductase is an important enzyme involved in the glyoxylate metabolism. It has been found in various organisms, including archaea, bacteria, yeast, algae, plants and human. For the plant, glyoxylate reductase helps to keep plant cells healthy and detoxifies the cell by reducing glyoxylate levels. In human, glyoxylate reductase/hydroxypyruvate reductase (hGRHPR) functionally converts the metabolic byproduct glyoxylate into the less reactive glycolate. In biopharmaceutical engineering, glyoxylate reductase also plays an important role, for it provides an opportunity for the synthesis of chiral hydroxy acids, which are of biotechnological interest as an intermediate for the synthesis of various pharmaceuticals.

Glyoxylate reductase belongs to the d-2-hydroxyacid dehydrogenase family. This enzyme catalyzes the reduction of glyoxylate to glycolate. Many glyoxylate reductases also catalyze the interconversion between hydroxypyruvate and d-glycerate. It can use both NADPH and NADH as a cofactor, but prefers NADPH. The oxidation of NADPH can be monitored spectrophotometrically at 340 nm.
