Enzyme Activity Measurement of Glucose 1-Dehydrogenase [NAD(P)+] Using Spectrophotometric Assays

Glucose 1-dehydrogenase [NAD(P)+] (EC 1.1.1.47) belongs to the family of oxidoreductases, it catalyzes the oxidation reaction of β-D-glucose to produce D-glucono-1,5-lactone, using NAD or NADP as the cofactor. Studies have shown that this enzyme has similar activity levels using either NAD+ or NADP+. This enzyme is also known as:

  • D-glucose dehydrogenase (NAD(P)+);
  • hexose phosphate dehydrogenase;
  • beta-D-glucose: NAD(P)+ 1-oxidoreductase;
  • glucose 1-dehydrogenase

Glucose 1-dehydrogenase [NAD(P)+] is related to the Entner-Doudoroff pathway. This pathway exists in some obligate anaerobic microorganisms, taking the role of the glycolysis pathway to generate pyruvate. Glucose 1-dehydrogenase [NAD(P)+] is the first enzyme in this pathway, catalyzing the oxidation of glucose into D-glucono-1,5-lactone. However, it has been shown that this enzyme has activity with a broad range of sugar substrates, including glucose, galactose, xylose, and L-arabinose, with specific requirement for the stereo configuration at the C2 and C3 positions of the sugar substrate. EC 1.1.1.47 from Bacillus has been deeply studied. It is a tetrameric enzyme with four identical subunits. The enzyme is inactivated in alkaline solutions, and the inactivation is due to the reversible dissociation of the tetramer into inactive protomers. Other sources of glucose 1-dehydrogenase [NAD(P)+] are found belonging to the medium chain alcohol dehydrogenase (MDR) superfamily, which needs zinc ions during the catalysis progress. The activity of these enzymes can be determined spectrophotometrically by following the increase in absorbance at 340 nm, corresponding to the reduction of NAD or NADP.
