Galactan 1,3-galactosidase (EC 3.2.1.145) belongs to glycoside hydrolase family 43 (GH43) and has been characterized from several sources, including bacteria and fungi. This enzyme particularly hydrolyzes the beta-1,3-galactan backbone of type II arabinogalactan (AG-II), bypassing beta-1,6-galactan side chains, and consequently releases galactose, beta-1,6-galactooligosaccharides and their derivatives. Arabinogalactans (AGs) are widely distributed in higher plants and can be grouped into two structural types, AG I and AG II. AG proteins (AGPs) have been implicated in many physiological events in plants such as differentiation, cell–cell recognition, embryogenesis, and programmed cell death. Type II AGs are generally linked to polypeptides, which are rich in hydroxyproline, serine, threonine, and alanine, forming AGPs. However, it is difficult to elucidate the relationships of the structures and functions of AGPs due to the structural complexities. Enzymes that are capable of degrading specific sites in AGs are valuable tools for determining their fine structures. Therefore, more and more studies are focused on galactan 1,3-galactosidase , targeting at structural identification and improving catalytic activity. Detecting the increase in reducing sugar is the most common way to assay the enzyme activity.
