dTMP kinase (TMPK, EC 2.7.4.9) catalyzes the phosphorylation of dTMP to dTDP, which is an essential step in the biosynthesis of nucleotide dTTP. This enzyme is a magnesium-dependent enzyme and prefers ATP as the phosphate donor. dTMP kinase is found in all organisms requiring the de novo pyrimidine synthesis. Cellular production of dTTP is a highly regulated process that is coordinated with DNA replication in the cell cycle. Therefore, this enzyme has a central role in the control of the dTTP pool size in eukaryotic cells. In addition to its vital role in supplying precursors for DNA synthesis, human TMPK has a significant medical perspective, participating in the activation of a number of pro-drugs, including HIV and herpes simplex virus infection treatment drugs. Meanwhile, due to the low sequence similarity with the human enzyme, TMPK from Mycobacterium tuberculosis has become a potential target for the development of new anti-tuberculosis drugs. The inhibitory effect is reflected by enzymatic activity, so a proper assay method for dTMP kinase is the vital part in enzymology studies. Several assay methods were developed targeting activity measurement of this enzyme. Nonetheless, to support getting more precise results, Creative Enzymes uses customized spectrophotometric methods by coupling the formation of ADP to the oxidation of NADH in reactions catalyzed by pyruvate kinase and lactate dehydrogenase, respectively.
