Delta4-3-oxosteroid 5beta-reductase is a soluble monomeric NADPH dependent enzyme and a member of the aldo-keto reductase (AKR) superfamily. Due to the essential role in primary bile acid and steroid hormone biosynthesis, delta4-3-oxosteroid 5beta-reductase has been well studied in mammals, especially in humans, where the enzyme is also designated as AKR1D1. AKR1D1 catalyzes the irreversible reduction of Δ4-3-ketosteroids to 5β-dihydrosteroids to introduce a 90° bend at the steroid A/B ring junction, using NADPH as the hydride donor.
