6-Phospho-beta-galactosidase (PGALase, EC 3.2.1.85) was first discovered in Staphylococcus aureus and the same catalytic activity was later demonstrated in Lactococci and Lactobacilli. According to the CAZy database, PGALase is assigned to GH family 1, which comprises glycosidases from organisms as far apart as archaea and man. PAGLases are enzymes that hydrolyze the beta-glycosidic bond between a terminal nonreducing galactose-6-phosphate and other organic molecules.
PGALase participates in vital metabolism activities. This enzyme is usually associated with the phosphotransferase system, which is also referred to as phosphoenolpyruvate-carbohydrate phosphotransferase system (PEP-PTS). This unique transport system is only found in bacteria. PEP-PTS is an alternative intake system involved in the transport and phosphorylation of numerous monosaccharides, disaccharides, amino sugars, polyols, and a series of other sugar derivatives. PGALase is involved in the utilization of lactose. Lactose is transported into the cell, phosphorylated via PEP-PTS system, and further hydrolyzed inside the cell by PGALase, resulting in glucose and galactose-6-phosphate. Up till now, only a small number of these enzymes have been biochemically characterized, among them, only PGALase from L. lactis has been subjected to full crystallographic analysis. Therefore, more and more studies are focused on the structural and the functional exploration in PEP-PTS.
