Tyrosinase (EC 1.14.18.1; L-tyrosine,L-dopa:oxygen oxidoreductase) is a member of the type III copper protein family harboring a catalytic center formed by dinuclear copper, which is able to perform two successive reactions in the presence of molecular oxygen: the hydroxylation of phenols to form ortho-diphenols (monophenolase activity), and the oxidation of o-diphenols to o-quinones (diphenolase activity). Tyrosinase is mainly used for melanin formation, which is responsible for skin pigmentation and the browning of fruit and vegetables. The enzyme can also serve as a component in wound healing and primary immune response. Tyrosinase is ubiquitous in almost all domains of life where it is involved in various biological functions and the active site of this enzyme is well conserved among the different species. The active site is typically formed of six conserved histidine residues which coordinate with two copper ions, denoted CuA and CuB.
Tyrosinase has great potential for the production of various o-diphenols (also known as substituted catechols), which are important intermediates for the synthesis of agrochemicals, flavors, polymerization inhibitors, and antioxidants. In mammals, this enzyme is related to the skin pigmentation abnormalities, such as flecks and albinism.
