Phospholipase C (PLC, EC 3.1.4.3) mainly exists in bacteria, catalyzing the hydrolysis of phospholipids to produce 1,2-diacyl-sn-glycerol (DAG) and phosphocholine. PLC is a zinc protein and it also acts on sphingomyelin and phosphatidylinositol.
Both PLC and its products have vital positions in growth and metabolism. In mammalian systems, the product DAG acts as a secondary messenger in a single transduction cascade by serving as an endogenous activator of protein kinase C. Besides, elucidation of the mechanisms responsible for aberrant phosphatidylcholine metabolism in cancer cells may allow identification of novel biomarkers of tumor progression and design of new targeted anticancer therapies.
