Phospholipase C (PLC, EC 3.1.4.3) mainly exists in bacteria, catalyzing the hydrolysis of phospholipids to produce 1,2-diacyl-sn-glycerol (DAG) and phosphocholine. PLC is a zinc protein and it also acts on sphingomyelin and phosphatidylinositol.

Both PLC and its products have vital positions in growth and metabolism. In mammalian systems, the product DAG acts as a secondary messenger in a single transduction cascade by serving as an endogenous activator of protein kinase C. In prokaryotes, PLC may serve as part of the phosphate retrieval pathway. In addition, PLC participates in other pathways such as:

  • ether lipid metabolism;
  • glycerophospholipid metabolism;
  • Inositol phosphate metabolism;
  • plasmalogen biosynthesis and degradation.

The important functions of phospholipase C have attracted increasing research interest. It is considered that PLC is one of the key enzymes for the design of drug delivery system using the endocytosis route because PLC can catalyze the membrane fusion between cell membranes and phospholipid vehicles (liposomes). Besides, elucidation of the mechanisms responsible for aberrant phosphatidylcholine metabolism in cancer cells may allow identification of novel biomarkers of tumor progression and design of new targeted anticancer therapies.
