Enzyme Activity Measurement for L-Lysine Oxidase

Unsurpassed by any others, Creative Enzymes provides innovative enzyme-related services, powers research with the most advanced instruments, and offers personalized technical support found nowhere else in the industry. Creative Enzymes is a leading company in the field of enzyme activity assays and we are proud to offer the rapid and accurate activity measurement for L-lysine oxidase.

L-lysine oxidase (EC 1.4.3.14) is a flavoenzyme that catalyzes the irreversible oxidative deamination of L-lysine to α-keto-ε-aminocaproate, concomitant with the production of ammonia and hydrogen peroxide. This enzyme was originally isolated from Trichoderma viride Y 244-2, and later it was also found in Trichoderma harzianum Rifai and some other marine organisms, such as the rockfish Sebastes schlegelii. Most of the L-amino acid oxidases show a wide range of substrate specificity, but not for L-lysine. However, this is not the case for L-lysine oxidase from T. viride, which exhibits a very high substrate specificity for L-lysine. This enzyme is stable over the wide range of pH and temperature, though the prosthetic flavin group could be released from the enzyme under acidic conditions. L-lysine oxidase is a member of the general oxidoreductase family, which catalyzes the CH-OH group of the substrate and uses NAD+ or NADP+ as the cofactor. The systematic name of this enzyme class is L-lysine:oxygen 2-oxidoreductase (deaminating), which is also known as L-lysine α-oxidase or L-lysyl-α-oxidase. L-lysine oxidase from T. viride is a homodimeric protein with an apparent molecular mass of 112 kDa. However, the molecular mechanisms of the enzymatic reaction and unusually high substrate specificity have not been well explored.

L-lysine oxidase from T. viride is useful in the food industry or fermentation process as an enzyme-based amino acid sensor for quantitative measurement of L-lysine concentration. Moreover, the enzyme has a strong antitumor activity and hence it is considered to be a potential anti-cancer agent. Furthermore, L-lysine oxidase plays a pivotal role in the biosynthesis of many value-added products including tropane, piperidine and pyridine alkaloid.

Owing to the multiple and critical functions performed by L-lysine oxidase, more detailed knowledge of this enzyme should be gained, especially through catalytic activity quantification. However, probably because of the unexplored working mechanisms of this enzyme, the activity assay of L-lysine oxidase seems to be difficult and multiplex. Fortunately, Creative Enzymes is the most experienced manufacturer specialized in enzyme activity assay development, which is supported by leading techniques, the best customer care and dedicated approach. Regardless of the challenges which may obstruct your way to enzyme applications, Creative Enzymes will always meet the requirements for the next development stage and will be your best choice for satisfying services.
