A glucuronolactone reductase (EC 1.1.1.20) is an enzyme that catalyzes oxidation of L-gulono-1,4-lactone, using NADP as the redox cofactor, to produce D-glucurono-3,6-lactone. The systematic name of this enzyme class is L-gulono-1,4-lactone:NADP+ 1-oxidoreductase, and other names in common use include GRase, and gulonolactone dehydrogenase. The enzyme belongs to the family of oxidoreductases. Glucuronolactone reductase also catalyzes the reversed reaction of D-glucurono-3,6-lactone with NADPH to produce L-gulono-1,4-lactone and NADP+. It was first isolated from the liver of Rattus norvegicus (rat) as a microsomal enzyme, and it was showed as the key catalyst in the biological pathway of ascorbate and aldarate metabolism.
In addition to the natural substrate, the enzyme also catalyzes reduction of D-glucuronic acid, which is a building block of proteoglycans and glycoglycerolipids, such as heparin, chondroitin, keratan, and hyaluronic acid. Regulations of glucuronic acid contents through reduction by glucuronolactone reductase would largely change functions of blood, skin, and joints. On the other aspect, the product of reducing D-glucuronic acid is L-gulonic acid, which is found useful in chemical processing.
