Glucuronoarabinoxylan endo-1,4-beta-xylanase (EC 3.2.1.136; glucuronoarabinoxylan 4-beta-D-xylanohydrolase) is an enzyme that catalyzes the endo-hydrolysis of (1,4)-β-D-xylosyl links in some glucuronoarabinoxylans. In addition, this enzyme presents a high activity towards feruloylated arabinoxylans from cereal plant cell walls. Recent studies have shown that glucuronoarabinoxylan endo-1,4-beta-xylanase from the Gram-positive bacterium Bacillus subtilis is from glycosyl hydrolases (GH) family 5. The major interest on this enzyme is the specificity and mode of action on the complex substrate glucuronoxylan and the characterization of the xylooligomeric products generated. The enzyme is glucuronoxylan-specific and cleaves the β-1,4-linked main xylan chain in an endo manner directed by the position of an α-1,2-linked glucuronate moiety. Besides, many reports have made a conclusion that cleavage happens at the second β-1,4 linkage toward the reducing terminus from a glucuronate-substituted xylose. This mode of action leads to signally substituted glucurono-xylooligosaccharides with the substitution penultimate to the reducing terminus. Functionally, glucuronoarabinoxylan endo-1,4-beta-xylanase serves as a critical role in the pre-treatment for ethanol formation from lignin-cellulose fibers. Furthermore, this enzyme can also be exploited in the food industry, such as in the fermentation and storing.
