Enzyme Activity Measurement for Gluconate 5-Dehydrogenase Using Spectrophotometric Assays

Gluconate 5-dehydrogenase (EC1.1.1.69; Ga5DH) is an enzyme that catalyzes a reversible oxidoreduction reaction between D-gluconate and 5-keto-D-gluconate, while simultaneously generating NADPH, which acts as a hydrogen donor for many biosynthetic processes. To date, biochemical characterization has been reported for gluconate 5-dehydrogenases from several species, including Gluconobacter oxydans, Gluconobacter liquefaciens, and Acetobacter suboxydans. The enzyme belongs to the short chain dehydrogenase/reductase (SDR) family with a preference for NADP+ rather than NAD+ as cofactor. The systematic name of this enzyme class is D-gluconate: NAD(P)+ 5-oxidoreductase. Other names in common use include:

  • 5-keto-D-gluconate 5-reductase;
  • 5-ketogluconate 5-reductase;
  • 5-ketogluconate reductase;
  • 5-keto-D-gluconate reductase

Gluconate 5-dehydrogenase plays an important role in L-idonate degradation, which ictions which are catalyzed by gluconate 5-dehydrogenase and another enzyme. D-Gluconate is then phosphorylated, forming 6-phosphogluconate which is an intermediate of central carbon metabolism. 6-Phosphogluconate is later metabolized via the pentose phosphate or Enter-Doudoroff pathway. Therefore, accompanied with energy consumption, 5-keto-D-gluconate can be converted to D-gluconate by gluconate 5-dehydrogenaseand and subsequently enter the Entner-Doudoroff pathway to join as a sous a critical pathway for ketogluconate metabolism. Gluconate 5-dehydrogenase is the second enzyme in the pathway of L-idonate degradation. In this pathway L-idonate is catabolized through the intermediate D-gluconate. L-Idonate is converted to D-gluconate by two consecutive oxidation and reduction rearce of carbon and NADP+. However, no structural information for gluconate 5-dehydrogenase is currently available to allow elucidation of the molecular architecture or clarification of the catalytic mechanism of the enzyme. Recently, some studies have shown that gluconate 5-dehydrogenase also seems like an effective target for the development of novel potent antibacterial agents. Despite of the multiple functions of gluconate 5-dehydrogenase in medical, biological, and pharmaceutical industries, the activity assay of this enzyme has not been well developed due to lack of structural information.
