Enzyme Activity Measurement for Fructuronate Reductase Using Spectrophotometric Assays

Fructuronate reductase (EC1.1.1.57) is an enzyme that catalyzes the reversible oxidation of D-mannonate to D-fructuronate in the presence of NAD+. And the enzyme also transforms D-tagaturonate to D-altronate. This enzyme is a member of the oxidoreductase which catalyze the CH-OH group of the substrate and use NAD+ or NADP+ as the cofactor. The systematic name of this enzyme class is D-mannonate:NAD+ 5-oxidoreductase, which is also known as mannonate oxidoreductase, mannonic dehydrogenase, D-mannonate dehydrogenase, and D-mannonate:NAD+ oxidoreductase. Fructuronate reductase is involved in multiple pathways:

  • D-fructuronate degradation;
  • Degradation of sugar acids;
  • Metabolic pathways;
  • Pentose and glucuronate interconversions

This enzyme is involved in the D-fructuronate degradation pathway, which is a critical part of the superpathway of β-D-glucuronide and D-glucuronate degradation. Escherichia coli can use β-D-glucuronides, D-glucuronate, or D-fructuronate as the sole carbon source for growth. The initial step in the degradation of β-glucuronides is hydrolysis to yield D-glucuronate, which is subsequently isomerized to D-fructuronate. D-fructuronate then undergoes an NADH-dependent reduction to D-mannonate catalyzed by fructuronate reductase. At this point, three pathways converged (the superpathway of β-D-glucuronide, D-glucuronate degradation, and D-galacturonate degradation I pathway). It is obvious that fructuronate reductase plays a critical role in bacterial growth and metabolism.
