Diacylglycerol kinases (ATP) (EC 2.7.1.107; DGKs) are a family of enzymes that catalyze the conversion of diacylglycerol (DAG) to phosphatidic acid (PA) using ATP as source of the phosphate. Diacylglycerol kinases (ATP) control intracellular signaling because both DAG and PA act as the second messengers. DAG activates protein kinase C (PKC) and has a wide range of targets, including the Ras guanyl nucleotiderelease proteins, chimaerins, and some transient receptor potential channels. Thus, the degradation of DAG by diacylglycerol kinases (ATP) can play a pivotal role in regulating various cellular processes. Furthermore, PA is involved in regulating multiple signaling proteins, including phosphatidylinositol 4-phosphate 5-kinase, PKC-ζ, and phospholipase C-γ1. Therefore, by attenuating DAG signaling and initiating PA signaling, the diacylglycerol kinases (ATP) can regulate multiple signaling events within the cell.

There are at least nine different isoforms of mammalian diacylglycerol kinases (ATP) which have been separated into five classes on the basis of their different structures and properties. All these isoforms have various critical functions in cellular activities. For instance, DGK-α, promotes cell proliferation, translocates to the plasma membrane, and is activated by calcium and by the lipid products of phosphatidylinositol-3-kinase. DGK-ε has been shown to regulate seizure susceptibility and long-term potentiation. Therefore, diacylglycerol kinases (ATP) are getting more and more attention because of their significance in multiple pathways.
