Enzyme Activity Measurement for Chloride Peroxidase

Chloride peroxidase (EC 1.11.1.10) is a group of enzymes that catalyze hydrogen peroxide-dependent halogenation reactions. By combining the inorganic substrates chloride and hydrogen peroxide, the organic substrate gets chlorinated. An example of the heme-thiolate chloride peroxidase is a unique fungal enzyme which is secreted by the ascomycetous fungus Caldariomyces fumago. In addition to catalyzing chlorination, bromination, and iodination reactions, chloride peroxidase is also considered to be a crucial model for the reaction intermediates of P450 enzymes that catalyze P450-like oxygen insertion reactions. Owing to this wide-ranging activity, chloride peroxidase is one of the most versatile of all known heme enzymes and has been the subject of extensive investigations since the enzyme was first described by Morris and Hager in 1966. Despite functional similarities with other heme enzymes, the structure of chloride peroxidase is unique, which folds into a tertiary structure dominated by eight helical segments. This enzyme belongs to the family of oxidoreductases, specifically those acting on a peroxide as acceptor (peroxidases). The systematic name of this enzyme class is chloride:hydrogen-peroxide oxidoreductase, which is also known as chloroperoxidase, CPO or vanadium haloperoxidase.

Chloride peroxidase plays a pivotal role in biotechnology, environmental protection, and biosynthesis. For example, chloride peroxidase can be used as a versatile biological catalyst because of its wide-ranging activity. Moreover, owing to the fact that chloride peroxidase shows oxidative dehalogenation activity and is significantly more robust than other peroxidases and functions under harsher reaction conditions compared to other biocatalysts, this enzyme can be used in the field of bioremediation and may be employed to treat contaminated soil or water prior to discharge. Furthermore, chloride peroxidase can also be used for the synthesis of semiconductor polymers. To this point, it should be seen that chloride peroxidase performs multiple and significant functions in many fields, thus much more details of this enzyme should be established, especially through its catalytic assay.
