Enzyme Activity Measurement for 6-Phospho-Beta-Glucosidase

6-Phospho-beta-glucosidase (EC 3.2.1.86; 6-phospho-beta-D-glucosyl-(1,4)-D-glucose glucohydrolase) belongs to glycoside hydrolase family 1 (GH1) and catalyzes the hydrolysis of β-1,4-linked cellobiose 6-phosphate (cellobiose-6’P) to yield glucose and glucose 6-phosphate, which is an important cellular precursor. The products of this reaction are further metabolized by the energy-generating glycolytic pathway. 6-Phospho-beta-glucosidase is a single-domain protein that adopts a (β/α)8-barrel structure, which is a typical fold of GH1 family members. According to the CAZy database, the GH1 family shows quite diverse enzyme functions and is composed of hydrolases with 19 enzymatic. 6-Phospho-beta-glucosidase from Thermotoga maritima is composed of a catalytic module of 415 amino acid residues and most likely exists in a dimer-tetramer equilibrium in solution. 6-Phospho-beta-glucosidase from T. maritima requires both NAD+ and Mn2+ for catalysis.
