TWS119, a 4,6-disubstituted pyrrolopyrimidine that can induce neurogenesis in murine ESCs. TWS119 can reduce beta-catenin phosphorylation and induce nuclear translocation of beta-catenin.TWS119 also elevate glutamine synthetase production, impede synthesis of alpha-smooth muscle actin and Wnt5a with promoting the expression of glial fibrillary acidic protein, Wnt10b, and paired-like homeodomain transcription factor 2c. Abeta(1-42) can be effectively prevented by treatment with the GSK-3beta inhibitor, TWS119, thereby implicating GSK-3beta kinase activity in this pathogenic cascade.
