In vitro
As an ATP competitive inhibitor, roscovitine can inhibit p33 cdk2/p35, p34 cdc2/cyclin B, p33 CDK2/cyclin A, and p33 CDK2/cyclin E with IC50 of 200 nM, 650 nM, 700 nM, and 700 nM. When combined with DNA damaging agents, R-Roscovitine was able to prevent the DNA damage-induced upregulation of cyclin A1 on a transcriptional and post-transcriptional level. Roscovitine can induce apoptosis in MCF-7 cell line. After roscovitine treatment, the level of wild-type p53 protein strongly increased and p53 was accumulated in the nucleus.
