1. Extract the tissue with guanidine-HCl to remove most of the proteoglycans.
2. Collagen was dissolved with pepsinized acetic acid at pH 3.0.
3. Fractional precipitation with NaCl and dialysis precipitation with Na2HPO4.
4. Column chromatography on CM-cellulose at pH 4.0.
5. Column chromatography on DEAE-cellulose at pH 7.5 to remove trace amounts of residual proteoglycans and collagen types I and III.
The proteoglycan content is less than 0.4%. Soluble in 0.01 M to 0.5 M HOAC.
